Peptide Identity vs. Purity: Two Different Analytical Questions

Identity and purity are often presented side by side, but they do not answer the same question. Responsible review keeps each result tied to its method, sample, lot, and stated limitation.

What an identity result addresses

An identity procedure is intended to distinguish the named material from other materials that might be confused with it. The evidence may come from mass, spectral, chromatographic, sequence-based, or other appropriately specific characteristics.

Specificity matters. FDA's Q6A guidance notes that a single chromatographic retention time alone is generally not considered a specific identification test. Combining evidence based on different principles can provide a more discriminating identity assessment.

What a purity result addresses

Purity is a measurement statement tied to a procedure. A chromatographic area percentage usually describes the relative detector response represented by the main peak under the stated conditions. It should not be silently reinterpreted as an absolute mass fraction or as proof that every possible impurity was detected.

Peptide purity assignment can require multiple measurement approaches and careful treatment of related impurities, water, counterions, residual solvents, and other components. NIST's peptide measurement work illustrates why a single number needs methodological context.

Assay, content, and amount are separate again

Assay or content is used to address the quantity of a defined component in a sample. The amount stated on a container is therefore not independently confirmed merely because a report includes a chromatographic purity percentage.

When amount matters to a research protocol, look for a procedure and result that actually address amount. Do not calculate an unstated content result by multiplying unrelated figures unless the method and reporting basis explicitly support that calculation.

Use an attribute matrix

A compact matrix prevents one result from doing the work of several. For each attribute, record the procedure, result, unit, limit, sample identifier, and date. Leave the cell blank when the report does not provide the information.

Identity
Which evidence differentiates the named material from plausible alternatives?
Purity
What was separated or detected, and how was the reported percentage calculated?
Content
Was the amount of the defined component measured with a suitable quantitative procedure?
Other attributes
Were water, residual solvents, counterions, bioburden, endotoxin, or other relevant characteristics evaluated?

Avoid the percentage shortcut

The safest reading is the narrow one: report exactly what the procedure measured and no more. A percentage without a named procedure, calculation basis, traceable sample, and supporting report is incomplete information.
